Spark Therapeutics, Inc., in Philadelphia, PA seeks a Senior Research Associate to: Work on Immunomonitoring Team to perform immunological assays to support novel gene therapy initiatives that challenge the inevitability of genetic disease. Responsible for conducting clinical and preclinical sample analysis, overseeing Immunomonitoring of nonclinical animal studies, including assay conceptualization, optimization, experimental design, planning, scheduling and project management, as well as performing the assays, analyzing data, compiling reports, and presenting data to the leaders of various therapeutic area teams.  Perform clinical sample ELISPOT and FluoroSpot analyses and report results to clinical teams. Prepare critical reagents, maintain equipment, train colleagues, lead the implementation of automation instrumentation in Immunomonitoring workflows, and act as a subject matter expert to author documents and collaboratively develop computer systems and upgrades.

In performing routine laboratory analyses, obtain clinical and preclinical samples through Laboratory Information Management Systems (LIMS) and prepare them for ELISPOT and FluoroSpot testing by thawing, washing, centrifuging, and enumerating peripheral blood mononuclear cells (PBMCs) to achieve a specific cellular concentration. Stimulate the PBMCs in specialized plate using peptide libraries. Following incubation, detect number of T cells that responded to stimuli mimicking thegene therapy by staining the specialized plate, perform image-based counting to enumerate the responding cells, and analyze data per standard operating procedures. Prepare reports for clinical and preclinical teams. Author standard operating procedures. Prepare critical reagents, ensure that equipment is properly maintained and functioning, and perform periodic logbook reviews per Good Laboratory Practice (GLP) requirements.

REQUIREMENTS: Master's degree in Biomedical Sciences, Biomedical Engineering, or related biological science field required plus knowledge of the following gained through post graduate coursework, employment, or internships: performing tissue culture,  ELISPOT OR FluoroSpot, and related cell-based immunoassays including ELISA OR electrochemiluminescence based assays (MSD); culturing human and animal cells in sterile biosafety cabinets; conceptualizing, optimizing, troubleshooting, and developing complex immunology assays; and performing PBMC isolation in animal or human whole blood.
