Position Summary

The Sr. Professional Research Assistant (Sr.PRA) will work within the School of Medicine, Division of Rheumatology (1.0 FTE). The PRA will join the research team of Isaac Harley, MD, PhD, MS. Dr. Harley’s research focuses on understanding the steps that lead to development of systemic lupus erythematosus (SLE or Lupus) and other autoimmune diseases. His studies investigate how genetic risk factors for autoimmune disease impact the autoreactive B cell and autoantibody responses that lead to disease. Specifically, his research focuses on the role of a microRNA that is a Lupus risk factor on the development of SLE and the biological role of other genetic risk factors in the development of this disease. To do this his lab uses a combination of big data approaches to define the relevant genetic risk variants as well as cell lines and primary cells from patients and murine disease models to understand the function of these risk genes and their role in the normal biology of immune cells.

Dr. Harley’s lab is embedded in the Departments of Immunology and Medicine. It is not expected that applicants will arrive with the full breadth of experience necessary for the job. It is expected that they will arrive with willingness to learn in areas where there is a gap in knowledge or experience. Importantly, there are lots of opportunities and a supportive environment that can provide training in any areas where there is a gap of knowledge or experience.

Work performed in this role will primarily consist of tissue culture experiments using human cell lines, human blood samples, mouse blood and tissue samples and Epstein-Barr virus infection to generate stable human lymphoblastoid cell lines. Additional responsibilities will include processing of human blood samples as well as using techniques such as flow cytometry, molecular biology and functional assays, such as RNA-Seq, qPCR, western blotting and ELISA. This position requires work in Biosafety Level (BSL2) facilities and will require development of proficiency in biocontainment procedures. Additional responsibilities will vary according to the active projects and the needs of the lab. These additional responsibilities are anticipated to include: recruiting for research studies, scheduling study participant visits, conducting study visits (including blood draw & urine collection), accurately recording and entering data, responding to data queries, supporting physician investigators, overseeing lab maintenance, laboratory data management, reagent ordering, maintaining inventory, adherence to biosafety regulations, submission of biosafety applications and training of new laboratory personnel. Minimum qualifications include a Master’s Degree or a Bachelor’s Degree and extensive prior biomedical laboratory-based research experience.

Most importantly, the successful candidate will be detail oriented and be able to work both independently and as part of a team. This includes being able to communicate appropriately and effectively with the principal investigator and other team members, possessing outstanding customer service skills, and maintaining relationships that are equally respectful to all. Research visits and study activities will be conducted at the Barbara Davis Center at the Anschutz Medical Campus in Aurora. On occasion, some study support activities will be completed remotely.

Examples of Work Performed

  • Processing of human blood, urine or tissue samples.

  • Cell culture experiments using human blood, urine or tissue samples.

  • virus infections to generate stable lymphoblastoid B cell lines.

  • Multicolor flow cytometry, spectral flow cytometry, RNA-Flow Cytometry, phosphor-flow cytometry [intracellular signaling], functional flow cytometry with live cells [Calcium flux assays]

  • Lentivirus/CRISPR-based genome-wide screening of B-cell receptor signaling pathways

  • Molecular biology (e.g. Molecular cloning, PCR)

  • Biological assays

  • Maintaining mouse colony (i.e. maintaining inventory, genotyping, animal husbandry)

  • Oversee lab maintenance and adherence to biosafety regulations, submission of biosafety applications

  • Ordering of reagents used in the lab and maintaining inventory

  • Training of new laboratory personnel.

  • Overseeing new laboratory personnel.

  • Review, develop and implement new or modified research protocols/standard operating procedures and trouble-shoot or problem-solve as related to the research project.

  • Prepare and present data during lab meetings and individual meetings with principal investigator

  • Review research protocol Inclusion/Exclusion Criteria and confirm eligibility of subject to participate in study. Perform informed consent process and/or ensures that the informed consent process has occurred, is properly documented, and that informed consent form documents are filed as required

  • Interview prospective subjects for a variety of research projects. Educates potential subjects on the details of the studies through phone contacts and personal interviews

  • Schedule subject participation in research studies, coordinating availability of necessary space and clinical research support

  • Independently conducts research study visits, to include the following:

  • Measurement of height, weight, and temperature

  • Phlebotomy (direct venipuncture)

  • Abstracts and records all research subject data pertaining to the research protocol in an accurate and timely fashion

  • Enters information for retrospective and observational studies into database

Salary and Benefits

The salary range (or hiring range) has been established at $49,000 - $60,000.

The salary of the finalist(s) selected for this role will be set based on a variety of factors, including but not limited to, internal equity, experience, education, specialty and training.

Qualifications

Minimum Requirements

Master’s Degree or equivalent combination of education and experience

(i.e. Bachelors degree in relevant field and 5 years or more of relevant experience).

Biomedical laboratory research experience is required.

Applicants must meet minimum qualifications at the time of hire.

Preferred Qualifications

– Ideal candidate will have 1 year or more experience with laboratory procedures and techniques relevant for working in a research lab focused on study of the impact of genetic variation on immune responses, such as flow cytometry, ELISA, molecular biology, CRISPR and/or biochemical methods.

– Knowledge of and experience with aseptic techniques to maintain sterility and culture cells.

– Experience in human subject research is preferred.

– Excellent organizational and time management skills to move projects forward are preferred.

– Excellent communication skills are preferred.

Comfortable with or willing to learn: processing human blood samples.

Comfortable with or willing to learn: experimental mouse colony maintenance and experiments with mouse models of human disease.

Additional Job Requirement: Flexible work hours, including infrequent evening or weekend hours may be required based on the need to process and store human blood and urine samples in a timely manner that will be suitable for future experiments.

Under the direction of the Principal Investigator (PI), the PRA will be assigned various research duties and will report to the relevant Principal Investigator (PI).

Supervision Exercised

The position may supervise student workers, rotating trainees (undergraduate students, graduate students and/or medical residents) or other lab members for specific tasks. It is anticipated that the clinical/translational research activities (phlebotomy, consenting, clinical sample processing, etc.) will be supervisory.
